One&Only Cape Town sits on Dock Road at the Victoria and Alfred Waterfront, divided between a crescent-shaped mainland building called the Marina Rise and a private island connected by waterways. The hotel holds one hundred and thirty-two rooms and suites, from sixty-three-square-metre Marina rooms with Table Mountain or harbour views up to the seven-hundred-and-ninety-eight-square-metre Lion's Head Penthouse with four bedrooms. Rooms in the Marina Rise have large private balconies; island rooms sit at the water's edge with marina views.

Four restaurants and bars operate on site. Nobu Cape Town is the only Nobu restaurant in Southern Africa, serving Japanese cuisine with Peruvian influence. ROOI is a South African grillroom cooking over fire. Isola serves Mediterranean and Capetonian food poolside. Vista Bar and Lounge occupies the lobby, with floor-to-ceiling windows facing Table Mountain, and serves afternoon tea and cocktails daily.

The spa sits on the hotel's island and uses ESPA products. Signature treatments include a rooibos and rock-salt scrub, a kelp wrap, and a warm poultice massage with local botanicals. The hotel also runs a supervised KidsOnly club for children aged four to twelve, a stand-up paddleboarding and kayaking operation beside the spa garden, and guided experiences that include yoga on Table Mountain, a tour of the Oranjezicht farmers market with the executive chef, and a sushi masterclass at Nobu.

The Two Oceans Aquarium is a short walk from the hotel entrance.
